What are the standards for electrophoretic processing?

The basic principle of electrophoresis is that it refers to the process of charged particles migrating under the action of an electric field. Many important biomolecules, such as amino acids, peptides, proteins, nucleotides, nucleic acids, etc., have ionizable groups that can carry positive or negative charges at a specific pH value. Under the action of an electric field, these charged molecules will move towards the electrode direction opposite to the polarity of their charge.

Electrophoretic skill is the ability to separate, determine or purify samples by utilizing the electric field, which causes charged molecules to move at different speeds due to the differences in the charged properties of various molecules in the sample to be separated, as well as the differences in the size, shape and other properties of the molecules themselves. The electrophoresis process needs to be carried out in a supporting medium. The free interface electrophoresis conducted by Tiselius et al. in 1937 did not have a fixed supporting medium, so dispersion and convection were relatively strong, affecting the separation effect. So it presents electrophoresis with a fixed supporting medium, where the sample undergoes electrophoresis process in a fixed medium, reducing the interference of dispersion and convection.

The initial supporting media were filter paper and cellulose acetate membrane, which are currently less commonly used in laboratories. For a long time, small molecule substances such as amino acids, peptides, sugars, etc. were usually separated and analyzed by electrophoresis using filter paper or cellulose or silica gel thin layer plates as media. However, currently, more active techniques such as HPLC are usually used for analysis. These media are suitable for separating small molecule substances, with simple and convenient operation. However, the separation effect of disordered biomolecules is relatively poor. The introduction of gel as a supporting medium has greatly promoted the development of electrophoresis skills, making electrophoresis skills become one of the important techniques for analyzing biological macromolecules such as proteins and nucleic acids.

Electrophoretic processing is not difficult, but we also need to know that there are certain standards for electrophoretic processing, otherwise we don't know if the quality is qualified. So what are the standards for this electrophoretic processing?

1. Smooth surface

This is a basic condition, if the surface is not smooth, then the basic qualification is lost;

2. Uniform thickness

Electrophoretic processing requires ensuring uniform thickness to prevent color differences from appearing;

3. No obvious cracks on the surface
